Enhancer‐mediated reporter gene expression in <i>Arabidopsis thaliana</i> : a forward genetic screen

چکیده

SUMMARY Gene expression is controlled and regulated by interactions between cis ‐regulatory DNA elements (CREs) regulatory proteins. Enhancers are one of the most important classes CREs in eukaryotes. Eukaryotic genes, especially those related to development or responses environmental cues, often multiple enhancers different tissues and/or at developmental stages. Remarkably, little known about molecular mechanisms which regulate gene plants. We identified a distal enhancer, CREβ , regulates AtDGK7 encodes diacylglycerol kinase Arabidopsis. developed transgenic line containing luciferase reporter ( LUC ) driven fused with minimal cauliflower mosaic virus (CaMV) 35S promoter. The enhancer was shown play role response osmotic pressure gene. A forward genetic screen pipeline based on established generate mutations associated altered suite mutants variable levels as well segregation patterns populations. demonstrate that this will allow us identify trans‐ factors function acting regulation endogenous

Facile high-throughput forward chemical genetic screening by in situ monitoring of glucuronidase-based reporter gene expression in Arabidopsis thaliana

The use of biologically active small molecules to perturb biological functions holds enormous potential for investigating complex signaling networks. However, in contrast to animal systems, the search for and application of chemical tools for basic discovery in the plant sciences, generally referred to as "chemical genetics," has only recently gained momentum. In addition to cultured cells, the...

Reporter gene expression for monitoring gene transfer.

The use of reporters such as green fluorescent protein (GFP) and firefly luciferase permit highly sensitive and nondestructive monitoring of gene transfer and expression. Modifications in GFP which increase intensity and thermostability, as well as alter its spectral qualities, have facilitated the use of GFP in a variety of gene transfer methods. Improvements in imaging technologies and their ...

Investigation of an Optimized Context for the Expression of GFP as a Reporter Gene in Chlamydomonas Reinhardtii

Background: Chlamydomonas reinhardtii is a novel recombinant eukaryotic expression system with many advantages including fast growth rate, rapid scalability, absence of human pathogens and the ability to fold and assemble complex proteins accurately, however, obstacle relatively low expression level necessitates optimizing foreign gene expression in this system. The Green Fluorescent Protein (G...

A New Reporter Gene Technology: Opportunities and Perspectives

The paper summarizes the current status of the reporter gene technology and their basics. Reporter gene technology is widely used to monitor cellular events associated with gene expression and signal transduction. Based upon the splicing of transcriptional control elements to a variety of reporter genes, it “reports” the effects of a cascade of signaling events on gene expression inside cells. ...

A green fluorescent protein reporter genetic screen that identifies modifiers of Hox gene function in the Drosophila embryo.

Hox genes encode evolutionarily conserved transcription factors that play fundamental roles in the organization of the animal body plan. Molecular studies emphasize that unidentified genes contribute to the control of Hox activity. In this study, we describe a genetic screen designed to identify functions required for the control of the wingless (wg) and empty spiracles (ems) target genes by th...
